Khavinson Bioregulators (Epitalon, Pinealon)

Navigation

1. Pharmacodynamics & Mechanism of Action

Developed at the St. Petersburg Institute of Bioregulation and Gerontology by Prof. Vladimir Khavinson, peptide bioregulators are ultra-short di-, tri-, and tetrapeptides ( amino acids, molecular weight ) isolated from or modeled on tissue-specific polypeptide extracts (Cytomedines).

  • Epigenetic Chromatin Decondensation & Promoter Binding: Because ultra-short peptides are smaller than the pore diameter of the nuclear pore complex, they enter both the cytoplasm and the cell nucleus via PepT1/PepT2 proton-coupled oligopeptide transporters. Biophysical modeling and circular dichroism studies indicate that specific tri/tetrapeptides bind complementary motifs in the minor groove of double-stranded DNA and interact with linker Histone H1 and core Histone H4—inducing localized heterochromatin-to-euchromatin unfolding (decondensation) in senescent cells and restoring tissue-specific gene transcription.
  • Epitalon / Epithalon (Ala-Glu-Asp-Gly / AEDG): Synthetic tetrapeptide modeled on the bovine pineal gland polypeptide extract Epithalamin.
    • Telomerase (hTERT) Induction: Induces the expression of the catalytic subunit of human Telomerase Reverse Transcriptase (hTERT) in telomerase-negative human somatic fibroblasts, elongating telomeres by an average of and allowing cells to bypass the Hayflick limit (continuing past 34 population doublings to doublings without malignant transformation).
    • Pineal Circadian Restoration: Upregulates pineal Arylalkylamine N-Acetyltransferase (AANAT) and pCREB transcription, restoring nocturnal melatonin synthesis and normalizing flattened diurnal cortisol rhythms in aged primates and humans.
  • Pinealon (Glu-Asp-Arg / EDR): Synthetic neuroprotective tripeptide targeting the cerebral cortex.
    • Neuronal Anti-Apoptosis & Dendritic Spine Preservation: Suppresses neuronal reactive oxygen species (ROS) accumulation, inhibits Caspase-3 activation, activates the MAPK/ERK pro-survival cascade, and upregulates Tryptophan Hydroxylase-2 (TPH2) ( synthesis) and Irisin (FNDC5) gene expression.
  • Companion Tissue-Specific Bioregulators:
    • Thymalin / Vilon (Lys-Glu / KE): Thymic immune rejuvenation, T-cell differentiation, and euchromatin activation.
    • Cortagen (Ala-Glu-Asp-Pro / AEDP): Cerebral cortex & peripheral nerve regeneration (modeled on Cortexin).
    • Vesugen (Lys-Glu-Asp / KED) & Ventfort: Vascular endothelial homeostasis and Ki-67 endothelial renewal.
    • Bronchogen (Ala-Glu-Asp-Leu / AEDL) & Cardiogen (Ala-Glu-Asp-Arg / AEDR): Bronchial and myocardial fibroblast remodeling.

2. Pharmacokinetics & Transport Mechanics

  • PepT1 / PepT2 Transporter Entry: Di- and tripeptides (like Vilon and Pinealon) and select tetrapeptides are absorbed intact across intestinal enterocytes, the blood-brain barrier, and cellular/nuclear membranes via SLC15A1 (PepT1) and SLC15A2 (PepT2) oligopeptide transporters without requiring endocytic receptor internalization.
  • Dosing Discrepancy Note (Epitalon vs. Epithalamin): A frequent biohacking error is confusing the dosing of crude bovine pineal extract Epithalamin ( IM, which contains active short peptides by weight) with pure synthetic Epitalon (AEDG). In Khavinson’s comparative primate and cell studies, pure Epitalon is active at lower concentrations ( SubQ or up to for 10 days), making daily protocols unnecessarily high.

3. Dosing, Cycling & Synergies

  • Standard Protocol (Biannual Pulse Protocol):
    • Epitalon (Synthetic AEDG): SubQ (or intranasally) for 10 to 20 consecutive days, repeated twice per year (every 6 months). Administer in the evening ( before bed) or morning if initial circadian phase-shifting temporarily increases dream intensity.
    • Pinealon (Synthetic EDR): SubQ/intranasal (or oral capsules) for 10 to 20 consecutive days, pulsed 2–3 times per year.
    • Synergistic Pineal-Thymic Longevity Pulse: Pair Epitalon () with Thymalin ( polypeptide or Vilon dipeptide) for 10 days twice yearly (mirroring the 12-to-15-year human mortality trials).
  • Contraindications & Biomarkers to Monitor:
    • Precautions: While Khavinson’s rodent bioassays showed Epitalon decreased spontaneous tumor incidence (via melatonin/immune surveillance restoration), caution is advised with any telomerase activator in active oncology.
    • Biomarkers: 6-Sulfatoxymelatonin (aMT6s) in first-morning urine (direct index of nocturnal pineal melatonin output), 4-Point Diurnal Cortisol, Lymphocyte Subset Panel ( ratio), and Leukocyte Telomere Length (qPCR / Flow-FISH) + Epigenetic GrimAge/DunedinPACE clocks.

4. Key Scientific Evidence Summary

  • Human Somatic Telomerase & Telomere Elongation: Khavinson et al. (2003) and Anisimov et al. demonstrated in Bulletin of Experimental Biology and Medicine that Epitalon induces telomerase activity in telomerase-negative human fetal fibroblasts (line 602/17), elongating telomeres by and extending replicative lifespan past the 34-division Hayflick limit to divisions.
  • Primate & Elderly Human Melatonin Restoration: Korkushko et al. (2006) and Goncharova et al. (2005) showed that Epitalon/Epithalamin restores nocturnal plasma melatonin by in aged rhesus monkeys and elderly humans while normalizing pancreatic insulin response.
  • Long-Term Human Mortality Reduction (Epithalamin + Thymalin): Korkushko et al. and Khavinson & Morozov (2003) reported in a 6-to-12-year randomized prospective clinical study in elderly cardiovascular patients () that biannual courses of Epithalamin and Thymalin reduced all-cause mortality by (and up to in combined pineal+thymic cohorts).
