Neurotrophins (BDNF, NGF, Lion’s Mane)

Navigation

1. Pharmacodynamics & Mechanism of Action

Neurotrophins—principally Brain-Derived Neurotrophic Factor (BDNF) and Nerve Growth Factor (NGF)—govern adult hippocampal neurogenesis (subgranular zone of the dentate gyrus), dendritic arborization, axonal myelination, and long-term synaptic consolidation. Because native BDNF ( homodimer) and NGF cannot cross the blood-brain barrier (BBB), biohacking interventions utilize BBB-permeable small molecules that either stimulate endogenous astroglial neurotrophin transcription or act as direct TrkA/TrkB receptor agonists.

  • Lion’s Mane (Hericium erinaceus — Hericenones & Erinacines):
    • Hericenones (A–H, from fruiting body) and NDPIH / Hericene A: Stimulate astroglial NGF and BDNF signaling cascades (ERK1/2 CREB).
    • Erinacines (A–I, specifically Erinacine A from liquid-fermented mycelium): Low-molecular-weight cyathane diterpenoids () that readily cross the BBB, potently upregulating hippocampal and locus coeruleus NGF and BDNF synthesis, promoting oligodendrocyte myelination, and reducing plaque burden.
  • Direct TrkB Agonists (7,8-DHF & 4’-DMA-7,8-DHF): 7,8-Dihydroxyflavone (7,8-DHF) binds the extracellular domain of the TrkB receptor with high affinity (), provoking receptor dimerization and autophosphorylation at to activate downstream PI3K/Akt, MAPK/ERK, and PLC- pathways—mimicking BDNF without activating pro-apoptotic receptors. Its synthetic derivative 4’-DMA-7,8-DHF (Eutropoflavin) exhibits higher potency and prolonged duration.
  • Bacopa monnieri (Bacosides A & B) & Polygala tenuifolia (Tenuigenin / Yuanzhi-1):
    • Bacopa monnieri: Upregulates hippocampal BDNF, enhances Tryptophan Hydroxylase (TPH2) and serotonin transporter (SERT) expression, increases dendritic branch points in the basolateral amygdala and hippocampus over 6–12 weeks, and upregulates endogenous SOD/catalase/glutathione peroxidase.
    • Polygala tenuifolia (BT-11): Triple monoamine reuptake inhibitor (DAT/NET/SERT), NMDA antagonist, and potent BDNF/TrkB inducer that prevents stress-induced hippocampal atrophy.

3. Dosing, Cycling & Synergies

  • Standard Protocol:
    • Lion’s Mane Dual-Extract: (standardized for -glucans + verified Erinacine A / Hericenones) in the morning.
    • Bacopa monnieri: Synapsa (CDRI-08) (less serotonergic lethargy) or Bacognize (taken in the evening to leverage sleep support) for at least 12 consecutive weeks.
    • 7,8-DHF: sublingually (or 4’-DMA-7,8-DHF early AM; cycle 5 days on / 2 days off to prevent TrkB receptor internalization/ubiquitination).
  • Synergies: Paul Stamets / Neurogenesis Stack (Lion’s Mane + Niacin/flush B3 for peripheral microvascular vasodilation), Zone 2 / HIIT aerobic exercise (releases myokine Irisin / FNDC5 and -hydroxybutyrate / Lactate which deacetylate the BDNF promoter IV), and Omega-3 & Lipid Membrane Health.
  • Contraindications & Biomarkers to Monitor:
    • Precautions: Lion’s Mane contains compounds that mildly inhibit 5-reductase and acts as a -opioid receptor (KOR) modulator (in Erinacine E); discontinue if libido reduction or dysphoria occurs. NGF elevation sensitizes peripheral TRPV1 and mast cell receptors—caution in histamine intolerance or chronic pruritus/neuropathic hyperalgesia. Bacopa inhibits acetylcholinesterase and can increase GI motility if taken fasted.
    • Biomarkers: Serum BDNF, DHT / Free Testosterone (if monitoring 5-reductase sensitivity), and validated delayed word-recall (RAVLT) scores.

4. Key Scientific Evidence Summary

  • Lion’s Mane Human MCI Trial: Mori et al. (2009) demonstrated in a 16-week double-blind, placebo-controlled RCT () that Hericium erinaceus significantly increased cognitive function scores at weeks 8, 12, and 16 (), with scores declining 4 weeks after discontinuation.
  • Discovery of 7,8-DHF as a TrkB Agonist: Jang et al. (2010) published in PNAS that 7,8-DHF binds TrkB with nanomolar affinity, crosses the BBB, induces TrkB phosphorylation in the rodent brain, and rescues neurogenesis and synaptic plasticity in BDNF-deficient models.
  • Bacopa Memory Consolidation: Stough et al. (2001) and Kongkeaw et al. (2014, meta-analysis of 9 RCTs, ) confirmed that 12 weeks of Bacopa monnieri () significantly improves speed of visual information processing, learning rate, and delayed memory consolidation ().
