Tissue Repair Peptides (BPC-157, TB-500, GHK-Cu)

Navigation

1. Pharmacodynamics & Mechanism of Action

Regenerative signaling peptides accelerate the transition from acute inflammation to proliferative angiogenesis, tenocyte/myoblast migration, and type I/III collagen organization.

  • BPC-157 (Body Protection Compound-157; GEPPPGKPADDAGLV): Synthetic pentadecapeptide derived from human gastric juice that remains stable in gastric acid for .
    • FAK-Paxillin & GH Receptor Upregulation: Activates Focal Adhesion Kinase (FAK) and paxillin phosphorylation in tendon fibroblasts, accelerating tenocyte outgrowth and upregulating Growth Hormone Receptor (GHR) expression on healing fibroblasts by up to (synergizing with GH Secretagogues (Ipamorelin, CJC-1295, Tesamorelin)).
    • VEGFR2 & eNOS Homeostasis: Stimulates VEGFR2 internalization and Akt-eNOS activation to drive collateral angiogenesis in avascular tissues (tendons, ligaments, menisci) while protecting the gastrointestinal mucosa and tight junctions (ZO-1, Claudin-1).
  • TB-500 / Thymosin (Ac-SDKP active N-terminal domain / 43-AA parent):
    • G-Actin Sequestration & Cytoskeletal Migration: Binds monomeric G-actin ( stoichiometry), preventing premature polymerization until endothelial, keratinocyte, and satellite stem cells migrate into the wound bed.
    • Anti-Fibrotic & Cardiac Repair: Downregulates myofibroblast -smooth muscle actin (-SMA) to prevent scar fibrosis while releasing the anti-fibrotic tetrapeptide Ac-SDKP.
  • GHK-Cu (Glycyl-L-Histidyl-L-Lysine-): Endogenous human plasma tripeptide ( at age 20, dropping to by age 60) with high copper(II) affinity. Modulates human genes ( up/down shift in Broad Institute Connectivity Map), resetting senescent fibroblast transcriptomes, balancing MMPs and TIMPs, and stimulating Type I/III collagen, elastin, glycosaminoglycans, and nerve outgrowth.
  • KPV (Lys-Pro-Val): C-terminal tripeptide of -MSH; enters intestinal epithelial and immune cells via PepT1 transporters to inhibit NF-B nuclear translocation and mast cell degranulation without causing melanogenesis.

2. Pharmacokinetics & Route Selection

PeptideMolecular WeightOptimal RouteSystemic vs. Local ActionStability Profile
BPC-157 (Acetate vs. Arginate)SubQ (musculoskeletal) or Oral Arginate (GI lumen)Systemic (SubQ reaches all tissues); Arginate salt shows gastric stabilityExceptionally stable in human gastric juice ()
TB-500 / Thymosin (TB4) / fragmentSubQHighly systemic (low molecular weight allows rapid interstitial diffusion)Requires bacteriostatic water reconstitution & storage
GHK-CuSubQ (systemic) or Topical/Microneedling (dermal/follicular)Both systemic ECM remodeling and local dermal collagen synthesisHigh stability; SubQ injection can cause transient copper-ion site soreness
KPVOral capsule / SubQ / IntranasalAbsorbed intact via intestinal PepT1 di/tripeptide transporterStable orally for IBD / gut barrier protocols

3. Dosing, Cycling & Synergies

  • Standard Protocol (“The Wolverine Stack”):
    • BPC-157: SubQ twice daily (or oral Arginate salt twice daily for GI barrier repair).
    • TB-500 (Thymosin ): SubQ twice weekly (or daily micro-dosing).
    • GHK-Cu: SubQ (diluted with extra bacteriostatic water or co-drawn with BPC-157 to eliminate injection-site sting) + topical serum.
  • Cycling Cadence: 4 to 8 weeks on, followed by at least 4 weeks off.
  • Synergies:
    • Collagen + Vitamin C Loading: Ingest hydrolyzed collagen peptides (Amino Acids & Precursors) + Vitamin C prior to isometric/eccentric tendon loading while on BPC-157/TB-500.
    • Zinc Balance: Pair systemic GHK-Cu with Zinc Picolinate/Bisglycinate (at a separate time of day) to maintain optimal serum ratio ( to ).
  • Contraindications & Biomarkers to Monitor:
    • Contraindications: Active malignancy or history of angiogenesis-sensitive tumors (pathological VEGF and actin polymerization pathways can theoretically promote tumor neovascularization); proliferating diabetic retinopathy; WADA-tested athletes (BPC-157 and TB-500 are prohibited under WADA S0/S2).
    • Biomarkers: hs-CRP, ESR, Serum Copper & Ceruloplasmin, Plasma Zinc, and VEGF (if monitoring angiogenesis).

4. Key Scientific Evidence Summary

  • Tendon Fibroblast Migration & FAK-Paxillin: Chang et al. (2011) demonstrated in Journal of Applied Physiology that BPC-157 dose-dependently accelerated tendon explant outgrowth, enhanced cell survival under oxidative stress, and activated FAK-paxillin phosphorylation.
  • ** Tendon GH Receptor Upregulation:** Chang et al. (2014) published in Molecules that BPC-157 increased Growth Hormone Receptor (GHR) mRNA and protein expression on tendon fibroblasts by up to 7-fold, potentiating GH-induced proliferation.
  • GHK-Cu Transcriptomic Reset: Pickart & Margolina (2018) detailed Broad Institute Connectivity Map data showing GHK-Cu modulates of the human genome ( expression shift in genes), suppressing metastatic/inflammatory genes while upregulating DNA repair and extracellular matrix remodeling.
